You have to match the higher pitched response to the lower pitched notes that are played. You do this by toggling the circles on the left, which are played in a loop from top to bottom. The dash in the vertical line indicates which circle is going to be played next. (Explanation thanks to Liam :D)

I do think, though, that with a tutorial this could really be something! :)
If you want to make the game more challenging, there could be a "survival" mode in which the player must face randomly selected sequences of notes and act fast, and see how much time he/she can survive! But I really suggest some faster way to assign notes to the markers on the left, maybe using the arrows to select which marker, and the keys 1 to 4 to select which note to play.
The graphics aren't impressive, but it really doesn't matter much in this kind of game. You could keep with this project in a educational context, it seems perfect to train the musical ear!

Nice concept! After a bit of work given to make it easier to learn what to do exactly and with some differences between levels this could be really good. :)
I love the piano sample played on completing the level. :D
